Magnesium glycinate vs oxide vs citrate: which one for the gut?

Most magnesium supplements are the wrong form for the job. The form decides the effect, not the milligrams on the label.
Walk down the supplement aisle and every bottle says magnesium. That word alone tells you almost nothing about what is in your hand. Magnesium is a metal. It only exists in a supplement bound to something else, and that something else changes everything. How much your body absorbs. Whether it calms your nervous system or sends you to the bathroom. Whether it earns its space in your stack or wastes the slot.
The TL;DR before the details. Glycinate for nerves and sleep. Citrate for constipation. Oxide for almost nothing. The form matters more than the dose.
Why the word magnesium alone is meaningless
Pure magnesium is reactive and unstable. To put it in a capsule, manufacturers bind it to a partner molecule and form a salt. That partner determines two things at once. How well the molecule dissolves in your gut. And how much of the magnesium actually crosses into your bloodstream rather than staying in your intestine.
Schuette and colleagues (1994) compared magnesium oxide to magnesium citrate in a controlled absorption study and found roughly four times higher absorption with citrate. Walker and colleagues (2003) ran a similar comparison and showed organic forms like citrate and amino acid chelates outperformed inorganic forms like oxide by a wide margin. Form is not a marketing detail. Form is the entire mechanism.
This is why two products with the same milligram number on the label can produce wildly different outcomes. One feeds your tissues. The other passes through and pulls water with it.
The four forms you actually encounter
Most shelves carry the same handful of forms in rotation. Here is what each is good at and where each fails.

What oxide actually does in your colon
Magnesium oxide is the cheap default. It is the form you find in most generic multivitamins and in bulk magnesium tablets at the drugstore. It is cheap because it is dense with magnesium by weight and easy to manufacture. None of that matters if your body cannot absorb it.
What does not get absorbed stays in the gut. Magnesium is osmotically active, meaning it pulls water with it. Sit a poorly absorbed magnesium salt in your small intestine and colon, and water follows. The stool softens. Then it loosens. Then it runs. This is exactly how magnesium oxide and magnesium hydroxide work as over-the-counter laxatives. Milk of Magnesia is magnesium hydroxide for this reason.
So when a multivitamin lists 250 milligrams of magnesium oxide and you wonder why the supplement gives you loose stools but does nothing for your sleep or muscle tension, you have your answer. The dose on the label is not the dose in your bloodstream.
The magnesium-glycine bonus
Glycinate is magnesium bound to glycine, an amino acid. The reason this form gets recommended for sleep and nerves is not just that magnesium itself is calming. Glycine is doing work in parallel.
Glycine is an inhibitory neurotransmitter in the central nervous system. Bannai and Kawai (2012) reviewed glycine's role in sleep and found that supplemental glycine before bed improved sleep quality and reduced daytime fatigue in human studies. It quiets nervous system signaling on its own. So a magnesium glycinate capsule delivers two calming inputs in one molecule, plus the amino acid chelate format keeps both of them well absorbed.
For people targeting muscle tension, racing thoughts, or trouble falling asleep, this combination matters. You are not paying for magnesium alone. You are paying for magnesium plus a co-absorbed calming amino acid in the same dose.
Magnesium L-threonate hype vs reality
L-threonate is the form sold on the claim that it crosses the blood-brain barrier and raises brain magnesium specifically. The marketing is built almost entirely on one paper. Slutsky and colleagues (2010) showed that L-threonate raised brain magnesium and improved learning and memory in rats. That is a real finding. It is also rats.
Human data is limited. A small trial in older adults with subjective cognitive complaints (Liu 2016) showed some cognitive improvement, but the study was small, industry-funded, and not replicated at scale. The form is also expensive per dose of elemental magnesium, meaning you pay more for less.
It is not a scam. It is just oversold. If cognition is your specific target and budget is not a constraint, it is a reasonable experiment. If you want general magnesium repletion or calming effects, glycinate gives you more for less.
The dosing reality
The recommended daily allowance for adult magnesium runs between 310 and 420 milligrams of elemental magnesium per day. Most adults in surveyed populations come in below that. Diet is the main lever: dark leafy greens, nuts, seeds, legumes, and whole grains. A supplement closes the gap. It is not meant to be heroic.
When you read a label, look for the elemental magnesium number. A 1000 milligram magnesium glycinate capsule is not 1000 milligrams of magnesium. The glycine partner takes up most of the weight. The actual elemental dose is closer to 100 to 200 milligrams depending on the format. Reputable labels list both.
Mauskop's 2012 review of magnesium for migraine prophylaxis used doses in the 400 to 600 milligram elemental range and found consistent benefit across multiple trials. That sits at the upper end of normal supplemental dosing and is well tolerated when the form is right. Go higher only with a clinical reason and a clinician's input.
What to do
- Read the form on the label, not just the milligram number. The salt is the active variable.
- If your goal is calm, sleep, or muscle tension relief, choose glycinate. Look for elemental magnesium in the 100 to 300 milligram range per serving.
- If your goal is occasional constipation relief, citrate is the right tool. Use it deliberately, not daily, unless directed by a clinician.
- If a current supplement is causing loose stools, check the form. Oxide or hydroxide are the usual suspects. Switch forms, lower the dose, or both.
- Take magnesium with food and in the evening when targeting sleep. Split larger doses into morning and night for better tolerance.
The bottle that costs four dollars and the bottle that costs forty can both say magnesium. Only one of them is doing the work you actually wanted.
